Still getting acquainted w/ my 2000 GT. Understanding that we didnt buy these cars for economy but still looking for every edge. The question is; considering the shift characteristics and overall behavior of the 4R70W, in town, would this car get any better economy with the OD turned off?

I'm not sure about the over drive but how many mpg would you say you are getting thru town as it is?
I'm getting about 13-15 mpg in my cobra with with my lead foot.
I've heard that loosing the spare tire and keeping a good eye on your tire pressure you should notice a difference in gas mileage.
Also just make sure your car is running smoothly.
Just throwing some stuff out there

I get 16-18 in my sixxer, sooo yours doesn't seem terrible. I would expect better hwy mpgs however. Probably closer to 25 or so.
