A tuner by itself is the most cost effective, because you can change the tunes are delivered based on your car's current configuration. So, if your car is stock, a tune can be made for regular or premium fuel, firmer/softer shifts, customized shift points, etc. Even better, as you mod your car, you can get new tunes that accommodate your new mods.
For the money, a tuner is the absolute best thing you can by for your car, even BEFORE a CAI.

Everything state except the shift points (obviously). Each tune is different, but it can change things like cam timing, engine timing, air/fuel mixture, and other engine operating parameters. A 87-octane street tune can give better performance AND better mileage, for instance. I have an automatic, so it also improves the quality of the shifting of the transmission. A tune can also be configured to disable the speed limiter (but I wouldn't recommend doing this unless you've replaced the drive shaft).

Exactly what joey said. Bama is great for all around use or beginner tuning. If you go all hardcore on us youll have to pay more and go to a place and get it dyno'd like brenspeed or jon lund
